Description: The use of triphenylphosphonium (TPP+)-conjugated bioactive molecules in mitochondrial biology was reinvented and refined by Murphy and coworkers. TPP+ cations were utilized to deliver the probes, antioxidants, and pharmacophores to mitochondria. The advantages of TPP+-based mitochondrial targeting over other approaches for mitochondrial delivery of small molecules include the stability of the TPP+ moiety in biological systems, a combination of lipophilic and hydrophilic property, the relatively simple synthesis and purification, the low chemical reactivity toward cellular components, and their lack of light absorption or fluorescence in the visible or near infrared (NIR) spectral region.
